Abstract d -β-Hydroxybutyrate dehydrogenase (BDH) (EC 1.1.1.30), a membrane enzyme, has been purified to homogeneity from dromedary ( Camelus dromedarius ) liver mitochondria. Our new purification method consisted of the solubilization of mitochondrial membranes by Triton X 100 and purification of BDH by two steps: DEAE-Sephacel and Phenyl-Sepharose. The molecular mass of the enzyme subunit size was 67 kDa. The purified enzyme is recognized by anti rat liver mitochondrial BDH antibodies. Furthermore, BDH activity was absolutely dependent upon phospholipids. The microenvironment characteristics — polarity, viscosity and order degree - in Triton X-100 reverse micelles (RM) in cyclohexane and benzene — n-hexane 30 : 70 (v/v), were investigated with the aid of a homologous series of cationic spin probes, CAT n, of 5- and 16-doxyl stearic acids and of a new absorption probe, 4-nitropyridine-N-oxide (NP). The spectral parameters were related to local hydration values by means of a series of poly(oxyethylene) (PEO)/water calibration mixtures. Spectrophotometric determination of mercury(II) and silver(I) with copper(II) and diethyldithiocarbamate in the presence of triton X-100

Abstract Procedures for the determination of mercury and silver by displacement of diethyldithiocarbamate (DDTC) from its copper complex in the presence of 1% Triton X-100, and measurement of the decrease in the Cu(DDTC) 2 absorbance, are described. The use of the surfactant avoids the need for an extraction step. Reproducibility within 1% and detection limits of 0.25 ppm Hg(II) and 0.45 ppm Ag(I) have been obtained, and linear calibration ranges up to 13 ppm Hg(II) and 15 ppm Ag(I). In the presence of 0.1 M EDTA very good selectivity is achieved.
Time-dependent monomerization of bacteriorhodopsin in triton X-100 solutions analyzed by high-performance liquid chromatography

Abstract Bacteriorhodopsin from Halobacterium halobium was monomerized in Triton X-100 solutions. The process of delipidation was monitored by size-exclusion high-performance liquid chromatography under conditions that preserved the native conformation of the protein. The effects on the process of monomerization of the concentration and pH of the Triton X-100 solutions were investigated. The monomeric bacteriorhodopsin separated was active in light-dependent proton translocation when incorporated into soy bean lecithin liposomes.
Properties of the F0F1 ATPase Complex from Rhodospirillum rubrum Chromatophores, Solubilized by Triton X-100

1. A cold-stable oligomycin-sensitive F0F1 ATPase complex from chromatophores of Rhodospirillum rubrum FR 1 was solubilized by Triton X-100 and purified by gel filtration. 2. The F0F1 complex is resolved by sodium dodecyl sulfate electrophoresis into 14 polypeptides with approximate molecular weights in the range of 58000--6800; five of these polypeptides are derived from the F1 moiety of the complex which carries the catalytic centers of the enzyme. 3. The purified F0F1 complex is homogeneous according to analytical ultracentrifugation and isoelectric focusing. 4. The molecular weight as determined by gel filtration is about 480 000 +/- 30 000. Studies on the UV-photolytic decomposition of some tensides and complexones inhibiting the inverse-voltammetric determination of metal ions

The UV-photolytic decomposition of some tensides and complexones has been investigated using the DPASV peak of indium as an indicator. The results show that UV-irradiation alone can completely decompose these compounds at lower concentrations within a short time. Higher concentrations need an additional oxidising agent.
